SOP for Operation and Maintenance of Microscope

PURPOSE
To lay down a procedure for operating and maintaining the microscope.

SCOPE
This SOP applies to any person operating the microscope.

RESPONSIBILITY
QC Microbiologist in charge of handling the microscope.

PROCEDURE
  • Plug the power plug into an external power outlet (check if the external power supply voltage matches the input power supply voltage before plugging it in).
  • First, adjust the brightness adjustment handle to the minimum position, turn on the power switch, and turn the brightness adjustment handle to the center position. Models without brightness adjustment handles do not have this adjustment.
  • The specimen is placed on the stage, fixed by the leaf spring clip, or placed in the mechanical moving platform clip, and the hand wheel is moved in vertical and horizontal directions. Let the specimen enter the light path.
  • When using the Abbe condenser, move the condenser lift lever to raise or lower the condenser; when using the vertical lifting mechanism, push the lever to raise or lower the condenser to meet different magnifications. Objectives lens requirements. This adjustment is not required for models without a lifting concentrator. For models with mirrors, the mirror should be rotated to bring external illumination to the specimen. The concave mirror is suitable for large-area external illumination sources (such as bright sky or fluorescent lamps).
  • Turn the nosepiece, put the objective lens into the optical path, observe from the eyepiece, turn the coarse-motion focusing hand wheel in the course fretting structure, see the image contour, then turn the micro The dynamic focus hand wheel makes the object image clear; in the coarse micro motion coaxial structure, the coarse motion focus hand wheel is also rotated first, and then the fine movement focus hand wheel.
  • When using the eyepiece, first adjust the fixed eyepiece tube to make the image clear, then adjust the eyepiece vision circle to make the eyepiece image clear, then rotate the double tube to make it with the observer the eye distance is the same. Place the desired magnification objective lens into the optical path and adjust it repeatedly as described above until both eyes can observe a clear ob·ect ima eat the same time.
  • Rotate the light bar or turn the aperture light, the bar lever can change the aperture diaphragm size.
  • When using the pointer eyepiece, cooperate with moving the specimen, the pointer can be pointed to the key observation target for discussion by other observers.
  • When using the teaching eyepiece group, the same specimen can be observed simultaneously by two people from the two eyepiece tubes.
  • When the instrument automatically slides down after long-term use of the instrument, turn the adjustment ring slightly in the direction of the arrow. The coarse hand wheel is tightened to prevent the stage from sliding down.
  • Replace the bulb and fuse.
  • Turn off the power and unplug the power cord.
  • Loosen the chassis lower cover screw and remove the cover to replace the bulb and fuse according to various model requirements.
  • The cover should be restored and the screws tightened after replacing the bulb and fuse.

MAINTENANCE
  • The instrument should be stored in dry, cool, clean, isolated, and non-corrosive gases.
  • The instrument is strictly corrected for the factory, and shall not be disassembled and assembled.
  • The exposed lens surface shall not be hand touched, the dust and dirt can be washed in ears or soft brush removal. Dip a little xylene or alcohol ether (3:7) mixed fluid to wipe with a soft cloth when the microscope is necessary to clean.
  • If observed with objective 100X (oil), the cedar oil on the lens and specimen should be wiped with cotton dipped in benzene and alcohol ether (3:7) mixture after observing.
  • When using and transporting the microscope, the action should be gentle to avoid strong vibration and collision. Forbid makes the left and right sides of the rough micro-focusing hand wheel at the same time reverse rotation, to avoid damage to the focusing mechanism.
