SOP for Operation of Shimadzu UV–1900 UV Spectrophotometer

OBJECTIVE
To lay down a procedure for the operation of a UV Spectrophotometer using UV Probe software.

SCOPE
This is applicable for the operation of a UV Spectrophotometer using UV Probe software for Shimadzu UV 1900.

RESPONSIBILITY
It is the responsibility of QC Executives to follow the procedure without deviation.

PROCEDURE
  • Clean and check the instrument for power supply and plugin.
  • Switch ‘ON’ the instrument by pressing on the black switch situated on the right, bottom side of the instrument.
  • The instrument will initialize to check if the internal parameters are complying, then place it in PC mode.


  • Switch ‘ON’ the computer.
  • From the desktop click on an icon displayed as “LabSolutions” and the software will request a username and password.

  • After Sometime the software will ask for a change of password.
  • After the software opens, it will show the selected project, instrument type, selections of instruments, Postrun, administration, and manual.

  • Select the project and click on the instrument then open the icon “UV-1900i” and the “SHIMADZU LabSolutions UV-Vis” will display
  • From the display, Choose the type of mode, whether spectrum, photometric, Quantitation or time course.


Spectrum Mode
  • Spectrum mode is for wavelength scanning over a continuous wide range.
  • Enter your wavelength range and scan speed (Medium), and click on OK.
  • The maximum allowable wavelength range is 1100 nm to 190 nm.
  • Zero the baseline by clicking on the “Baseline” button near the bottom of the screen. Be sure that there are no samples in either the sample or the reference side while correcting the baseline.

  • “Auto zero” the baseline and fill the blank solution in the cuvettes and place it in the sample holder and press “START”, use the same producer on the sample and standard scans.



  • After the scan is completed save the file with Product Name/Test/Batch No. etc.
  • To take print out right-click and print.
  • After finishing all the scans saved and the file if no further work, click on “disconnect” and close the software and switch off the instrument and clean the cuvettes.


Photometric mode
  • Is for quantitative analysis, measuring absorbance at single wavelengths.
  • Remove anything left in the sample chamber, close the lid then press “Auto zero”. Fill the blank solution in the cuvettes, place it in the sample holder, and press “Auto zero”.
  • Fill the cuvettes with the standard solution or the sample solution, place it in the sample holder, and then read the absorbance.
  • After finishing all the scans save the file if no further work will be done click on “disconnect” and close the software and switch off the instrument and clean the cuvettes.
