SOP for Calibration of HPLC Column Oven

Objective
To provide a standard operating procedure for the calibration of the HPLC Column Oven.

Scope
This procedure is applicable to all HPLC Column Ovens in the QC Lab.

Procedure
Frequency: Once a year.


Calibration Procedure for HPLC Column Oven
  • Keep the sensor probe of reference calibrated digital thermometer inside the HPLC Column Oven and keep its position in about the center of the oven chamber.
  • Set the oven temp. at 25℃ .
  • Let the oven chamber achieve a set temp. Allow it to stabilize for at least 5 min.
  • After 5 minutes, observe the temp. in ref. thermometer.
  • Record the value of temp. shown on the instrument display and ref. the thermometer in Annexure–I.
  • Keep a minimum of five-minute intervals for the first and second readings.
  • Then set the oven temp. 40°C, 50°C, 60°C and 70°C each one by one and follow the procedure as mentioned in the above steps.
